Current fields of research

Project manager of the REGRESS study (Regression Growth Evaluation Statin Study), a multicenter placebo controlled study to assess the effects of 2 year treatment with pravastatin on pro- and regression of angiographically documented coronary artery disease in 885 patients with a serum cholesterol between 4 and 8 mmol/l, using quantitative coronary angiography and B-mode ultrasound.
Large substudies include specialised lipid research and evaluation of genetic markers of the dynamics of coronary atherosclerosis. Study sponsors: Bristol Myers Squibb and the Interuniversity Cardiology Institute of the Netherlands (ICIN).

Principal investigator of the cardiac branch of the "Pancreas Transplantation Study", an international multicenter study to assess the success of combined kidney and pancreas transplantation. The cardiac branch of this study evaluates the effects of the transplantation on progression and regression of coronary artery disease assessed by quantitative coronary angiography.

Principal investigator of several animal model studies (ApoE3 Leiden transgenic mice) for studying behaviour of atherosclerosis and responses of atherosclerosis to various drug regimens by means of histological and immunological techniques. Furthermore vascular wall tissue constituents are studied by Raman spectroscopy. These studies are carried out in close cooperation with the T.N.O.-PG Gaubius Laboratory (Prof. L.M. Havekes, PhD, H.M.G. Princen, PhD). Sponsors: Pfizer and Ministery of Economics.

Investigator of atherosclerosis research protocols using Raman spectroscopy, a new promising technique that enables quantitative analysis of the chemical composition of the vascular wall. By illuminating the tissue with infrared laser light and collecting the light emitted from the tissue, highly molecule specific Raman spectra can be obtained. Cooperation with the Erasmus University Rotterdam the Netherlands, and the Massachusetts Institute of Technology (MIT), Cambridge, Mass, and the Cleveland Clinic Foundation, Cleveland, Ohio.

Investigator of research protocols evaluating endothelium dependent vasodilation in the forearm of patients with different kinds of dyslipidemia on different treatment modalities.

Participating investigator in several intra coronary stent trials combined with intra coronary ultrasound, currently focussing on long term effects of stent coatings, both in humans as well as in transgenic atherosclerotic mice.

Investigator and Dutch coordinator of the TNT (Treat to New Targets) trial. An international multicenter double blind randomized trial, to study the effect of LDL-cholesterol lowering beyond currently recommended minimum targets on coronary heart disease recurrence in patients with pre-existing coronary heart disease. Patient are randomized to atorvastatin 10 mg or 80 mg. Clinical events will be evaluated in 8600 patients in five years follow-up time. This study is sponsored by Pfizer Int.

Investigator and member of the end point committee of the PROSPER trial (PROspective Study of Pravastatin in the Elderly at Risk). An international, three centre study, double blind randomized, to assess the efficacy of pravastatin to prevent stroke, myocardial infarction and cognitive impairment in 5500 elderly subjects. This study is sponsored by Bristol Myers Squibb Inc.

Investigator of a study to detect significant stenoses in saphenous bypass graft by magnetic resonance imaging (MRI), correlated to intra coronary Doppler flow and flow reserve assessments. This study is supported by the Netherlands Heart Foundation (grant 97.173).

Principal investigator a genetic studies which studies cholesteryl ester transfer protein (CETP) gene variation; an important factor in the response to lipid lowering therapy. This study is sponsored by the Netherlands Heart Foundation (grant 1998.163)

Principal investigator of the Genetic Determinants of Restenosis (GENDER) project. A multicenter study to investigate genetic aspects of restenosis after PTCA. This study is supported by a grants of the Interuniversity Cardiology Institute of the Netherlands (ICIN) and the Netherlands Heart Foundation (grant 99.210 ). The project has started in Feb. 1999.

Investigator of Statin in Diabetics (LiDIA) study. A Dutch study to investigate the effects of cerivastatin on vascular morbidity, evaluated by forearm blood flow and 24 h ECG monitoring, in 250 type-2 diabetic patients.
This study is sponsored by Bayer AG/MSD.

Main investigator of the Prevention of re-Infarction with early treatment by Cerivastatin Study (PRINCESS). An international randomized placebo controlled study with cerivastatin early or 3 months after myocardial infarction.

Principle investigator of the comparison of Rosuvastatin and Atorvastatin in different Dosages and effects on Reversed cholesterol transport (RADAR) study. A Dutch study in 600 patients.

Principal investigator of the GREAT study "Genes of Restenosis Evaluated by Activity of Transcription ". Gene expression profiles of restenotic, atherosclerotic and normal vessel tissue are obtained by coronary atherectomy and compared. Gene expression profiles are generated using a custom-made micro-array including 4500cDNA clones. This study is sponsored by Guidant Inc.

Investigator on molecular imaging of corornary atherosclerosis (adhesion molecules, scavenger receptors).

Co-investigator in projects on modulation of gene-expression as therapy for atherosclerosis. Topics:
* New strategies (a.o. macrophage-specific photodynamic therapy) to eliminate (lipid laden) macrophages from the atherosclerotic plaque and evaluation of its therapeutic potential in atherosclerosis.
